ABSTRACT

Penicillium citrinum exposing chitinolytic activity was isolated from Can Gio mangrove forest. Conditions affecting the production of chitinases by Penicillium citrinum were optimised under solid substrate fermentation (SSF). The most suitable conditions for chitinase production by P. citrinum  were 10⁶ spore/g culture medium containing 10% chitin, 2% NaCl with pH 4.5 and moisture of 60-80%; the incubation  time was 36 hours at 40^oC. Penicillium citrinum yielded maximum chitinase 0.144 U/ml. Chitinase was partially purified with (NH₄)₂SO₄ showed optimum activity at temperature 60^oC and pH 6.0.

Chủng Penicillium citrinum có hoạt tính chitinase được phân lập từ rừng ngập mặn Cần Giờ. Các điều kiện nuôi cấy thích hợp cho sự sinh tổng hợp enzyme chitinase đã được tối ưu trong môi trường lên men bán rắn. Mật độ bào tử 10⁶ bào tử/g môi trường nuôi cấy có chứa 10% chitin, 2% NaCl, với pH 4,5 và ẩm độ ban đầu 60-80%, thời gian nuôi cấy 36 giờ ở  nhiệt độ 40^oC, là thích hợp nhất cho sự sinh tổng hợp chitinase của chủng nấm này. Hoạt tính chitinase cao nhất của chủng này là 0,144 U/ml. Dịch enzyme được tinh sạch sơ bộ bằng (NH₄)₂SO₄ có hoạt tính chitinase cao nhất ở nhiệt độ 60^oC và pH 6,0.
