This study examined the antioxidant and tyrosinase inhibitory activities of the extract from Portulaca oleracea L. in vitro. The plant sample was macerated and extracted in 90% ethanol. Liquid-liquid extraction was used for separating the crude ethanol extract by increasing-polarization solvents. The phytochemical analysis showed the presence of alkaloids, flavonoids, saponins, steroids, tannins and terpenoids in Portulaca oleracea L. extracts. The total phenolic and the total flavonoid contents were determined by Folin-Ciocalteu and aluminum chloride colorimetric method, respectively. Methanol extract was fragmented with the highest total phenolic content (13,2±0,52 mg GAE/g extract), the ethyl acetate extract was the highest in total flavonoid content (17,37±0,62 mg QE/g extract). The highest antioxidant effect showed on ethyl acetate fragment by DPPH assay (IC50 = 219,21±1,34 µg/mL). The tyrosinase inhibitory activity of the extracts from Portulaca oleracea L. was investigated by determining formation of dopachrome at 475 nm. The most tyrosinase inhibitory activity was examined by ethyl acetate extract (IC50 = 77,85±1,36 µg/mL). Base on these results, Portulaca oleracea L. showed antioxidant and tyrosinase inhibitory effects. This performance is probably more related to the flavonoid found in this plant.
