The effects of ubiquitin-proteasome system on the degradation of the cytoskeleton were cited in some previous studies, this phenomenon caused the drip loss formation in muscle cells decreasing the water holding capacity (WHC) of meat. However, the drip production is enhanced by the muscle contraction, which is promoted by the accumulation of Ca2+ relating to the porcine stress syndrome. In this study, we sough to evaluate the interaction between the polymorphisms of UBXN1 and TRIP12 genes, which belong to the ubiquitin-proteasome system, and the mutation of RYR1 gene, to illustrate that potential interaction might affect WHC of pork. The total ca. 330 pigs derived from commercial herds of Pietrain x (German Large White x German Landrace) were used for this study. The significant interaction between the variation of UBXN1 at locus c.674C>T (p.Thr225Ile) with the RYR1 genotypes was detected for muscle conductivity(P<0.05). The animals with double heterozygous `CT/Nn´ had significantly higher the least square means for muscle conductivity than other interaction genotypes. The interaction of RYR1 genotypes and the SNP c.4957A>C of TRIP12 exhibited the significant association with the indicators of WHC. The `AC/Nn´ genotype had significantly lower muscle conductivity (P<0.1) but obtained higher pH value (P<0.05) than other combinations of genotypes. Thus, the polymorphisms of two genes showed the interactions with RYR1 mutation that impacted on drip loss formation in the muscle cells. Consequently, our findings imply that the effects of RYR1 gene on the PSE phenomena in pork might be influenced by the ubiquitination process.
